I have a major leak at what looks to be the rear main bearing at the hydramatic transmission on my 1954 Kaiser Manhattan. Do I have to drop the trans to install a new seal or is it a two-piece seal that can be accessed by removing the engine oil pan and 'chasing' the old seal out and pushing the new seal in.?

It's a two piece seal. It can be changed without removing the trans. The factory shop manual will help with this job.
